Help I don't have context I don't get the punchline

The non-Kindred Spirits characters in this comic are from two of Liar-soft's other titles, Gyousatsu♥Shinsengumi (panel 1) and Hiragumo-chan (panel 2+3), which are both works of historical fiction based on Japanese historical periods. Both games feature a high number of female characters, with Shinsengumi in particular having its heroines be genderbent versions of real historical samurai, so you can probably get an idea of what the punchline means from there.
And no, neither of those games are yuri, unfortunately.
